The recommended dose of cocaine hydrochloride ranges from 40 mg to 160 mg, depending on the nasal surface area to be anesthetized and the procedure to be performed. ( 2.2)

There are a multitude of reasons why buying drug test kits is a good idea. Among them? Rotting flesh, a chemical form of AIDS, organ damage, strokes, and even death are a few of the potential effects caused by substances often used to cut cocaine. These days, manufacturers looking to stretch their profits by mixing in other ingredients are more common than ever. Advise patients to inform their health care professional if they are taking CNS stimulants, alpha-modifying agents, monoamine oxidase inhibitors, or tricyclic antidepressants [see Drug Interactions ( 7.1)]. Adverse reactions have occurred in infants ingesting cocaine through breastmilk, including vomiting, diarrhea, convulsions, hypertension, tachycardia, agitation and irritability. The long-term effects on infants exposed to cocaine through breast milk are unknown. There are no data on the effects of NUMBRINO on milk production.

7. Drug Interactions

Single dose administration of cocaine intravenously during organogenesis in mice produced cryptochidism, anophthalmia, exencephaly, and delayed ossification at 7.8 times the HRD. In rats, a single dose of cocaine administered by intraperitoneal injection produced edematous fetuses, hemorrhages and limb defects at 12.9 times the HRD ( see Data). Advise patients that cocaine hydrochloride may lower the seizure threshold. Patients should be monitored for the development of seizures [see Warnings and Precautions ( 5.2)]. One spray delivers 130µl of solution (containing 13mg of Cocaine). Therefore, a maximal dose of 1.5mg/Kg of cocaine (approx. 1ml of 10% solution), is equivalent to approximately 8-9 sprays for a 70Kg adult, and this dose must not be exceeded. Long-term studies in animals have not been performed to evaluate the carcinogenic potential of cocaine.

Cocaine is detected in human breastmilk in chronic abuse situations and is expected to be at higher concentrations in milk than in maternal blood based on its physicochemical characteristics. Breastfeeding immediately after administration of cocaine hydrochloride could result in infant plasma concentrations that are approximately half the anticipated maximum maternal plasma concentrations at the clinical dose of 160 mg. The effects of this cocaine plasma concentration in an infant are unknown, but no level of cocaine exposure is considered safe for a breastfed infant. Cholinesterase inhibitors: Concomitant administration may increase the risk of cocaine toxicity. ( 7.4) Marquis Reagent Test Kits: Primarily used to screen ecstasy, Marquis test kits look for MDMA-type ingredients, as well as other ingredients that are commonly used as adulterants. This test reacts to amphetamines, DXM, 2C-B, MDPV, and other ingredients. Cocaine is metabolised in humans by two major pathways. The first accounts for over 90% of the transformation and involves various hydrolytic reactions. The second is an oxidative process. The hydrolytic pathway is catalysed by serum and liver pseudocholinesterase which produces ecgonine methyl ester, benzoylecgonine and ecgonine. The minor oxidative route appears to be responsible for the hepatotoxicity. Cocaine is N-demethylated to produce norcocaine which is then rapidly oxidized to N-hydroxynorcocaine, which in turn is further metabolised producing norcocaine nitroxide. The latter is believed to be ultimately responsible for the hepatotoxicity elicited by cocaine . At clinical doses, cocaine has little general toxicity, when applied locally and for a short period of time.

Fake Cocaine: A Growing Threat

Some persons have a cocaine idiosyncrasy and death may occur quite suddenly after doses of only 20mg. Mandelin Test Kits: A Mandelin test kit will help you identify or rule out PMA, PMAA, amphetamines, MDxx, specific opiates, as well as 2C-type chemicals. The half life of cocaine in serum is dependent on the route of administration, and average values of 0.6 hour for intravenous administration, 0.9 hour for oral use, and 1.3 hours after intranasal have been quoted. However, these values must be interpreted with caution, as there is considerable variability between individuals and within individuals over time, and cocaine applied to the mucous membranes may be detectable in serum for as long as three hours after its application. labetalol, phentolamine or sodium nitroprusside (not propranolol since it potentiates cocaine toxicity - see below*)

Cocaine is taken from the coca plant, which grows in Peru, Argentina, Colombia, and other parts of South America. Chewing coca leaves for their stimulant and pain relief effects was the most common form of ingestion until cocaine was properly extracted and discovered in 1860. Avoid use of cocaine hydrochloride in patients with a history of myocardial infarction, coronary artery disease, congestive heart failure, irregular heart rhythm, abnormal ECG, or uncontrolled hypertension. Avoid use of additional vasoconstrictor agents such as epinephrine or phenylephrine with cocaine hydrochloride. If concomitant use is unavoidable, prolonged vital sign and ECG monitoring may be required.

Physical and Mental Effects

The efficacy endpoint was anesthetic success, defined as immediate anesthesia based on a score of 0 on a NPRS during a von Frey monofilament test 20 minutes after study drug administration, and sustained anesthesia throughout the diagnostic procedure or surgery based on no further anesthetic or analgesic treatment required. The apparent elimination half-life of cocaine following administration of cocaine hydrochloride (by pledgets) was 1.54 hours) for the 4% concentration. NUMBRINO should be administered by means of cotton or rayon applicator pledgets applied to the nasal mucosa. ( 2) Cocaine may cause restlessness, excitement, euphoria, garrulousness and increased motor activity. With high doses or repeated use, confusion, paranoia, hallucinations, altered tactile sensations and psychosis have been reported. Seizures can occur, perhaps due to lowering of the seizure threshold, or hyperpyrexia, or due to life threatening cardiac arrhythmias. Cocaine in any form can be habit-forming, leading to addiction and long-term damage to the nervous system, nose, mouth, and even your mental health.
